چکیده

Treatment with phenobarbital of rats which are genetically selected results in a greater than IO-fold increase in nicotinamide-adenine dinucleotide (NAD)-dependent aldehyde dehydrogenase present in the supernatant but not the mitochondrial fraction of liver. This effect represents one of only a few known cases where intrastrain differences in a drug-induced increase in enzyme activity is genetically controlled. The kinetic characteristics of the enzyme from phenobarbital-treated rats are the same, regardless of whether or not the enzyme activity in the liver is increased. It is also demonstrated that rat liver supernatant contains a second NAD-dependent aldehyde dehydrogenase which (a) is resistant to induction by phenobarbital; (b) oxidizes fi-carboxybenzaldehyde; (c) is relatively uninhibited by disulfirarn; (d) has a heat stability different from the inducible enzyme.

Participation of cytochrome P-450 in reductive metabolism of 1-nitropyrene by rat liver microsomes.

Reductive metabolism of carcinogenic 1-nitropyrene by rat liver microsomes and reconstituted cytochrome P-450 systems was investigated. Under the nitrogen atmosphere, 1-aminopyrene was the only detected metabolite of 1-nitropyrene. The reductase activity in liver 105,000 X g supernatant fraction was ascribed to DT-diaphorase, aldehyde oxidase, and other unknown enzyme(s) from the results of cof...

Phenobarbital-inducible aldehyde dehydrogenase in the rat. cDNA sequence and regulation of the mRNA by phenobarbital in responsive rats.

In the rat, a cytosolic isozyme of aldehyde dehydrogenase, designated ALDH-PB, can be induced in the liver by administration of phenobarbital (PB). ALDH-PB activity and mRNA are induced in Long-Evans rats that possess a responsive (R) allele but are not induced in homozygous nonresponsive rats (rr), although the rr genotype is competent to induce other PB-responsive mRNAs. ALDH-PB mRNA is expre...

Rat liver aldehyde dehydrogenases: strain differences in the response of the enzymes to phenobarbital treatment.

The effect of phenobarbital on activities of aldehyde dehydrogenases in the subcellular fractions was studied in vivo in rats of three strains. Regarding the responses of the mitochondrial and microsomal enzymes to phenobarbital treatment no difference among the strains was observed. Regarding the responses of the supernatant enzyme remarkable strain differences were observed. In rats of the Lo...

Purification of aldehyde dehydrogenase from rat liver mitochondria by alpha-cyanocinnamate affinity chromatography.

1. alpha-Cyano-4-hydroxycinnamate was coupled to Sepharose CL-4B activated with 1,2:3,4-bisepoxybutane. 2. The low-Km rat liver mitochondrial aldehyde dehydrogenase was specifically bound to this affinity medium, and could subsequently be eluted with alpha-cyano-4-hydroxycinnamate. 3. The enzyme purified in this manner had a subunit molecular mass of 55 kDa and a pI of approx. 6.5. A minor comp...

Effect of Phenobarbital on Chloramphonicol-Induced Toxicity in Rat Liver and Small Intestine

Objective(s): The aim of the present study is to determine the effect of Chloramphenicol (CAP) on rat liver and small intestine. Effect of phenobarbital (PB) on CAP toxicity was also investigated.   Materials and Methods:   Rats were received CAP at doses of 0, 200, 400 and 600 mg/kg. Another group was pretreated with 80 mg/kg PB 30 min prior to administration of various doses...
